Kaczmarek studierte an der Universität Wisconsin, wo sie auch Tony Shalhoub traf. Mit ihm war sie später Teil der Theatergruppe in Yale. Sie hatte viele erfolgreiche Auftritte am Broadway in New York. In Die Simpsons spricht sie die Richterin Constance Harm. Besondere Bekanntheit erreichte Kaczmarek auch außerhalb der USA mit der Sitcom Malcolm Mittendrin, in der sie die Rolle der matriarchalen Mutter Lois spielt.

Sie wurde sieben Mal (2000–2006) für den Emmy und 2001, 2002 und 2003 für den Golden Globe nominiert; außerdem 2001 und 2003 für die Screen Actors Guild Awards. Alle Nominierungen bezogen sich auf ihre Rolle als Lois in der Comedy-Serie Malcolm Mittendrin.

Jane Kaczmarek ist mit dem Schauspieler Bradley Whitford verheiratet. In der deutschen Fassung von Malcolm Mittendrin wird sie von Martina Treger und in Die Simpsons von Inge Solbrig gesprochen.
