Als Tochter eines Diplomaten wuchs sie bis zum College-Alter in Kanada, Kuba, Washington, Venezuela und England heran. Während sie in London zur High School ging besuchte sie regelmäßig Theateraufführungen und war von Bühnengrößen wie Diana Rigg, Christopher Plummer und Angela Lansbury so beeindruckt, dass sie Schauspielunterricht an der London Central School of Speech and Drama nahm. Nach dem Tod ihres Vaters kehrte die Familie in die USA zurück. Dort besuchte Kathleen Turner zunächst die Southwestern Missouri State University in Springfield und machte 1977 ihren Bachelor of Fine Arts an der University of Maryland Baltimore County.

Sie zog nach New York City, arbeitete als Kellnerin und trat in Off-Broadway-Produktionen auf. 1978 wurde sie für die langlebige Seifenoper The Doctors engagiert und nach nur einer Saison wieder entlassen. Bereits ihr freizügiges Leinwanddebüt in Lawrence Kasdans Film Body Heat bedeutete jedoch 1981 Kathleen Turners Durchbruch zum Filmstar. Wegen ihrer Schönheit, der markanten, rauchigen Stimme und des durch den Filmerfolg gewonnenen Images als klassische Femme fatale wurde sie mit Lauren Bacall verglichen.

Turner festigte jedoch ihren Status in Hollywood zunächst mit komödiantischen Rollen als Partnerin von Steve Martin in Der Mann mit den zwei Gehirnen (1982) und von Michael Douglas in der Indiana Jones-Parodie Auf der Jagd nach dem grünen Diamanten (1984). Ebenfalls 1984 war sie als Prostituierte wieder eine zwielichtige Heldin in Ken Russells China Blue bei Tag und Nacht. In John Hustons vorletztem Film Die Ehre der Prizzis (1985) spielte sie an der Seite von Jack Nicholson eine Killerin. In Danny DeVitos Der Rosenkrieg (1989) lieferte sie sich mit Michael Douglas einen mörderischen Ehestreit. In Francis Ford Coppolas Peggy Sue hat geheiratet (1986) hatten sich die Eheprobleme mit Partner Nicholas Cage noch durch eine Zeitreise gelöst. Für diese Darstellung einer erwachsenen Frau, die sich plötzlich zurück in der High School findet, erhielt sie eine Oscar-Nominierung.

Kathleen Turner war in den 1980er-Jahren durch ihre Darstellungen starker Frauenfiguren eine der erfolgreichsten Schauspielerinnen Hollywoods. Zu der Zeit schrieb der Pop-Sänger Falco auch den Song „The Kiss of Kathleen Turner“. Ihre Karriere wurde jedoch zunehmend von einer schweren Verlaufsform der rheumatoiden Arthritis und den mit der Medikation verbundenen körperlichen Veränderungen behindert. 1991 geriet die Sara Paretsky-Verfilmung V.I. Warshawski mit Kathleen Turner als zäher Privatdetektivin zum Flop. 1993 hatte sie ihren bislang letzten größeren Filmerfolg in der Titelrolle von John Waters schwarzer Komödie Serial Mom. 1999 ging Kathleen Turner wegen einer Alkoholabhängigkeit freiwillig in eine Entzugsklinik.

Sie trat in den letzten Jahren in zum Teil größeren zeitlichen Abständen als Charakterdarstellerin in Kino- und TV-Rollen auf und ist vor allem in Bühnenrollen erfolgreich. Am Broadway erntete sie zwei Tony Award-Nominierungen für Rollen, die Elizabeth Taylor im Film dargestellt hatte: 1990 für Die Katze auf dem heißen Blechdach und 2005 für Wer hat Angst vor Virginia Woolf?.

Sie ist seit 1984 in zweiter Ehe mit dem Grundstücksmakler Jay Weiss verheiratet. 1987 wurde ihre gemeinsame Tochter Rachel Ann geboren.
